真性赤血球増加症の原因遺伝子であるチロシンキナーゼJAK2の点変異体 (V617F)は、転写因子STAT5の活性化を介して細胞増殖や腫瘍形成を誘導する。本研究では、JAK2変異体の下流でSTAT5の活性化を介して、転写因子c-Mycおよびその標的遺伝子であるポリアミン合成酵素 ODCや分裂期キナーゼAurkaの発現が誘導されることを見出した。c-MycおよびODCはJAK2変異体による細胞増殖や腫瘍形成に寄与し、AurkaはJAK2変異体の持つ抗がん剤耐性に重要な役割を果たすこと明らかにした。
