本研究では、小型魚類であるメダカで細胞特異的にマウスの転写因子Fra-1 を発現させ、骨芽細胞におけるFra-1の機能を明らかにすることを目的とした。メダカで骨芽細胞特異的にFra-1 を発現するトランスジェニックメダカを作製したところ、骨石灰化の抑制が起こることが観察された。すなわち、Fra-1の発現を介してミネラリゼーションを抑制する分子機構が、マウスだけでなくメダカにもあることが示唆された。 We generated transgenic medaka fish expressing mouse Fra-1 in osteoblasts to determine cell-autonomous function of Fra-1 in osteoblasts in vivo. Alizarin complexone staining and micro-computed tomography revealed that Fra-1 transgenic medaka showed lower bone mineral density than wild-type controls. These data suggest that Fra-1 expression in osteoblasts impairs bone mineralization not only in mice but also in medaka.
