子宮体癌に対する術後補助化学療法の効果を早期に的確に判定する手法の構築を目指し、薬物反応を規定する宿主側因子としての一塩基多型(SNP)に着目した。網羅的SNP解析を行い、治療ターゲットとしての癌幹細胞個性診断を試みた。本研究により、化学療法時の薬物反応と相関する薬物代謝関連の特定のSNPを選別することができた。これらは化学療法の毒性の予測と同時に、治療効果をも予測する因子として有用であった。以上のことから、個体のゲノム情報に基づいた癌幹細胞個性診断が、新たな個別化治療戦略に応用できる可能性が示された。 Single nucleotide polymorphisms (SNPs) analysis is expected as the personalized medicine in pharmacotherapy. In this study, we attempted to extract SNPs correlated with drug response of adjuvant chemotherapy for uterine cancer as an individual diagnosis of cancer stem cells. A comprehensive SNP analysis was conducted, and specific SNPs on drug metabolism genes were identified. These SNPs may be useful in predicting toxicities as well as effects of chemotherapy. An individual diagnosis of cancer stem cells based on genomic information could apply to a new personalized treatment for uterine and other cancers.
