がん患者のセンチネルリンパ節(Sentinel lymph nodes : SLNs)における免疫抑制病態が抗腫瘍T細胞誘導に重要であるが, その構築機構は不明である。本研究は, がん細胞産生TGF-β1(Transforming growth factor beta 1)のがん免疫病態形成における意義を検討した。その結果, がん微小環境でのTGF-β1増加は, DC機能低下や免疫抑制性細胞増加により腫瘍組織とSLNsに免疫抑制状態を形成し, 最終的に抗腫瘍CD8+T細胞誘導や腫瘍浸潤の低下を起こす。従ってTGF-β1が, がん免疫抑制病態形成に強く関与し, がん治療効果増強の標的になることが示唆された。 In cancer patients, sentinel lymph nodes (SLNs) are crucial in the induction of antitumor T cells. However, in many cases, SLNs and tumors appear to be in immune-suppressive condition through mechanisms yet to be elucidated. In this study, the role of tumor-derived TGF-b1 in the generation of immunosuppressive microenvironments of tumors and SLNs was investigated. These results demonstrate that overproduction of TGF-b1 is critical for the generation of immunosuppressive microenvironments in both tumors and SLNs, which may result in suppression of spontaneous antitumor CD8+ T-cell responses. Therefore, TGF-b1 is an attractive target for restoration of immunosuppressive condition in cancer patients.
