本研究では胃がんにみられる遺伝子異常について網羅的に解析し, その生物学的特徴を解明するとともに薬効予測に有用な新規のバイオマーカーを探索することを目的として行った。胃がん細胞株を用いた実験の結果, CD44v8-10の発現とsulfasarazineの感受性の間に相関を認めた。さらに胃がん患者さんからの組織を用いた解析を追加し, HER2陽性胃癌に対する抗HER2抗体であるtrastuzumabの感受性に関連するバイオマーカーの探索を行い, 現在解析中である。 We tried to elucidate unknown biological characteristics in gastric cancer, and explore novel biomarker to predict drug sensitivity through the comprehensive genetic and biological analyses. We found correlation between expression level of CD44v8-10 and sensitivity of sulfasarazine in cell lines of gastric cancer. We currently analyze biomarkers to predict efficacy of trastuzumab, anti-HER2 antibody, in patients with HER2 positive gastric cancer.
