従来の免疫担当細胞のFACSや定量PCRによる解析手法では, 集団の「傾向」を見るにとどまり, 詳細なクローン組成や個々の細胞における定量情報は皆無であった。本研究では, DNAバーコード法を改良・発展させ, T細胞受容体ペアのハイスループットかつバイアスのない検出を行う方法の確立を試みた．本手法により, 免疫がダイナミックに活性化されている大腸炎モデルにおいて, 腸管浸潤T細胞のTCRレパトア解析を実施した。 Conventional methods such as flow cytometry and quantitative PCR have limitation to analyze individual lymphocyte cells. Here we established single cell analysis method by taking advantage of DNA barcoding, and analyzed TCR repertoire in the gut of experimental colitis model.
