生後4-6週の免疫不全ブタの肝実質内にヒト肝細胞癌株HepG2を超音波ガイド下で局注し, ヒト肝細胞癌ゼノグラフトモデルの作製に成功した。超音波検査で腫瘍の生着を確認した後に, 血管造影検査および造影CT検査を施行し, 早期濃染, washoutといったヒト肝細胞に矛盾しない画像所見が得られた。また病理学的に中-低分化型肝細胞癌に矛盾しない所見が得られた。今後は本モデルを使用した治療実験を行うことで, ヒト肝細胞癌に対する新たな治療モダリティの開発や, 現存治療の有用性評価などへの応用が期待される。 We succeeded the development of xenogragt model of hepatocellular carcinoma (HCC) by ultrasonography(US)-guided implantation of human HCC cell line "hepG2" into the liver parenchyma of immunodeficienct pigs. We demonstrated angiography and contrast CT scan after confirming the engraftment of tumor by US and early enhancement and washout were observed, which was compatible with human HCC. Furthermore, the engrafted tumor was pathologially compatible with poorly to moderately differentiated HCC. This model can be applied to develpment of novel treatment modality for human HCC or evaluation of efficacy of current treatments.
