研究成果として、巨大血小板の電子顕微鏡を用いた形態解析、遺伝子改変モデル動物を用いた機能解析の結果、国内外の本患者の表現型スペクトラムについて、研究分担者である上原朋子を筆頭著者、研究代表者 (武内俊樹) を責任連絡著者として、国際学会誌Scientific Reportsに英文論文として発表した。国内外の5人の患者について、詳細な表現型の比較検討を行った。その結果、巨大血小板性血小板減少症、知的障害、屈指、脳構造異常と感音性難聴、甲状腺機能低下症、易感染性が本疾患の核となる症状であることを見出した。患者由来の血小板を高解像度の電子顕微鏡で解析し、α顆粒は減少し、空胞が増加していることを見出した。 We performed electron micrographic analysis of the patients' platelets, and functional analysis of mutant C. elegans carrying the p.Tyr64Cys mutation in cdc-42. The results were published in Scientific Reports. Macrothrombocytopenia, intellectual disability, sensorineural deafness, distinctive facial features and structural brain abnormalities are cardinal features of Takenouchi-Kosaki syndrome.
