組織工学の分野では生体外で構築した三次元組織に毛細血管を導入する血管化を実現する手法が望まれている。本研究では、マイクロ流体デバイスおよびゲルビーズを用いることで三次元肝組織の血管化に取り組んだ。その結果、肝細胞の組織化にゲルビーズのサイズが重要であることがわかったが、血管化に対する効果としては限定的であった。また、間葉系幹細胞を用いると、血管網の安定化に寄与するだけでなく、血管化の促進にも重要な役割を果たすことがわかった。 In the field of tissue engineering, it is desired to introduce capillaries into reconstructed three-dimensional (3D) tissues in vitro, which is called vascularization. In this study, we investigated vascularization of tissue-engineered constructs by using microfluidic devices and gel microbeads. Our results demonstrated that the size of gel microbeads was important for tissue organization of hepatocytes while the effect of gel microbeads on vascularization was limited. In addition, we found that mesenchymal stem cells not only contributed to the stabilization of capillary networks but also promoted vascularization of tissue-engineered constructs.
