ヒト臨床検体を用いて50例の膵癌オルガノイドによって構成される膵癌オルガノイドライブラリーの構築に成功した。遺伝学的情報ならびに表現型解析を比較検討した結果、膵癌はWntとRspondinという２つの細胞増殖因子を生存に必要とするかどうかで、３つのサブタイプに分類可能であることが示された。すなわちWnt分非泌型膵癌、Wnt分泌型膵癌、Wnt/Rspo非依存型膵癌である。これらは段階的に悪性化してることが示唆され、表現型による膵癌の亜分類に成功した。 A library consisting of 50 human pancreatic tumor organoids was successfully established. Comparison between genetic information and phenotypic traits using the library revealed that pancreatic cancers were classified into three distinct subtypes based on Wnt or Rspondin dependence, namely Wnt non-secreting, Wnt secreting and Wnt/Rspondin independent type. This categorization suggested a gradual transition of pancreatic cancers to more malignant subtype along with the acquisition of Wnt Independence.
