本研究において、これまでに OATP1A2 および OATP2B1 発現 HEK293 細胞株を樹立し、その細胞膜上に発現する当該蛋白質を LC-MS/MS により定量することに成功した。OATP1A2 および OATP2B1 蛋白質発現量は、約 10～30 fmol 程度であった。これらと同時に内在性コントロール蛋白質として Na/K ATPase の発現量も測定した結果、既報と同等の発現量（約 50 fmol/mg protein) を確認することができた。一方、現在までにトランスポーターの絶対発現量を補正するためのマーカータンパク質発現細胞株の樹立に成功した。すなわち、当該マーカー蛋白質発現遺伝子を安定発現する細胞株を作成し、当該マーカータンパク質を蛍光標識することで、安定発現細胞株が樹立されたことが確認された。今後、当該発現細胞株を用いて Patch-clamp 法により、電流測定を行い、最終的に細胞膜上のトランスポーター発現量の補正法を構築する。 We are currently developing the novel methodology of quantification of absolute expression level of Transporter(s) such as OATP1A2 and OATP2B1, using QTAP and Patch-clamp method. For QTAP, we quantitatively analyzed the expression level of OATP1A2 and OATP2B1 on the plasma membrane using LC-MS/MS. The expression level of those transporters on the HEK293 cells were 10~30 fmol/mg protein. For Patch-clamp method, we now established the cell line which expressed Marker protein to adjust the expression level of OATPs.
