Note (General): ヒストンのメチル化修飾状態の制御異常は、がんの発症や悪性進展を引き起こす重要な要因のひとつと考えられている。我々は、ヒストン脱メチル化酵素のメンバーであるJMJD3遺伝子について、がんとの関連を調べるために、JMJD3ノックダウン細胞の表現型を解析した。JMJD3の発現を恒常的にノックダウンした細胞では、細胞運動能の亢進と上皮•間葉転換（EMT）の促進が観察された。遺伝子の発現解析から、E-CadherinなどEMTマーカー遺伝子やEMT誘導転写制御因子の発現変化にJMJD3が関与することがわかった。さらに、これらの遺伝子も含めて、JMJD3酵素が直接発現制御する標的遺伝子を探索するために、ChIP解析に使用するJMJD3特異的モノクローナル抗体の作製を現在進行している。
