長鎖ノンコーディングRNAのC型肝炎ウイルス（HCV)感染における役割を明らかにするため研究を実施した。HCV感染培養細胞のRNAを次世代シークエンサーで解析した所、長鎖ノンコーディングRNAの一つHULCの発現がHCV感染により増加することが明らかになった。HCV感染によるHULCの増加は、HCV感染チンパンジー、HCV感染マウスモデルでも認められた。またHCV感染培養肝細胞でHULCの発現を抑制したところHCV複製の低下を認め、この効果はHCVの翻訳過程の抑制を介していると考えられた。HULCは肝発癌の関与が示唆されており、HCVはHULCの発現を増加し肝発癌を誘発していると考えられた。
We investigated the role of long non-coding RNAs in HCV infection. The analysis on RNA of HCV-infected cultured hepatocytes by a next generation sequencer revealed that the RNA level of one of long non-coding RNAs, HULC, increased. The increase of HULC RNA level was observed not only in HCV-infected cultured hepatocytes, but also in an HCV-infected chimpanzee and a HCV-infected mouse. The knockdown of HCV by siRNA suppressed HCV replication as well as infectious virus production in HCV-infected cultured hepatocytes. This effect seemed to occur through the suppression of a translation step among HCV life cycles. Several recent reports show that HULC has the ability to promote hepatocarcinogenesis, therefore, the result of this study suggests a mechanism of hepatocarcinogesis due to HCV infection through HULC.
