Note (General): 本研究では、ヌクレオチド除去修復（nucleotide excision repair; NER）の必須因子ERCC1またはXPBを分解誘導する２種類の低分子化合物の作用機序を詳細に解析し、両反応に関わるユビキチンE3リガーゼ（SCF複合体）を各々同定した後、プロテインキナーゼの関与も明らかにした。また、NER阻害化合物を癌治療に応用する試みとして、シスプラチンの抗腫瘍効果に対する増強作用を示すこと、またオラパリブ（PARP阻害剤）との併用により相乗的な細胞障害性が得られることを示した。一方、新たな化合物ライブラリースクリーニングを行い、新規のNER阻害化合物を２種類同定することに成功した。
In this study, we have tried to uncover the detailed mechanisms underlying the protein degradation of nucleotide excision repair (NER) factors, ERCC1 or XPB, induced by two kinds of small-molecule compound. We have identified two different SCF complexes responsible for each reaction and also suggested the involvement of protein kinases. Futhermore, we have found that the ERCC1 degrader potentiates the cytotoxicity of cisplatin or olaparib to cancer cells. On the other hand, we have newly identified two natural compounds inhibiting NER with different mechanisms after another chemical library screening.
