本研究の目的は、任意の表現形質を担う遺伝子を機能面から系統的かつ簡便に同定する技術として代表者らが独自に考案したAntisense Display法を新しい機能性遺伝子スクリーニング技術として確立し、成人病性血管障害関連遺伝子、特に血管新生抑制性遺伝子の分離に適用することである。平成15年度の研究で、以下の成果を得た。1)平成13、14年度に行なった血管新生関連遺伝子のスクリーニングで同定された血管内皮細胞の増殖促進を示す14merアンチセンス(AS)配列を用いて、cDNA/ESTデータベースを検索し、パーフェクトマッチ配列を有する候補配列を5種同定した。2)ヒト血管内皮細胞での候補配列の発現をRT-PCR法とノーザン法で検討した結果、5種のうち1種、pICln遺伝子の発現が確認された。3)pICln mRNAの別領域に対するASの添加で、血管内皮細胞の増殖促進が再現され、さらに血管内皮細胞の管腔形成が促進された。4)AS処理細胞での各種血管新生関連遺伝子の発現を調べたところ、選択的スプライシングによって生じる2種の血管新生抑制性可溶型VEGF受容体(soluble Flt-1,soluble neuropilin-1)mRNAの発現が特異的に抑制されていることが示された。5)血管内皮細胞にcDNAを導入しpIClnを過剰発現させると、血管内皮細胞の増殖が有意に阻害された。さらに、pIClnを過剰発現細胞ではsoluble Flt-1,soluble neuropilin-1 mRNAの発現が特異的に増加していた。以上、今回同定されたpICln遺伝子は、選択的スプライシングを制御するというこれまで知られていなかった新しいタイプの血管新生抑制遺伝子である可能性が示唆された。
