Note (General): 金沢大学がん進展制御研究所
免疫抑制剤FK506によるNF-κB活性化の機序を明らかにするため、そのインヒビターのIκBαの分解について解析した。その結果、その分解は、IκBαのN末部の32番目と36番目のセリン残基(Seri32/Ser-36)に依存し、従来指摘されていたプロテアソームが関与していることを確かめた。さらに、Ser-32で燐酸化されたIκBαに対する特異抗体を用いてウェスタンブロットした結果、FK506がIκBαのSer-32の緩徐な燐酸化を引き起こすことが確認された。しかし、FK506はJNKの強い活性化を引き起こすにも関わらず、FK506によるIκB Kinase(IKK-1/-2)の活性化は認められなかった。また、IL-1によるIκBαの分解では、N末部のセリン残基の燐酸化に依存したN末部の21番目と22番目のリジン残基(Lys-21/Lys-22)へのユビキチン化が必須であるが、FK506によるIκBαの分解はこのN末部のユビキチン化部位に依存しないで起こった。以上、FK506によるIκBαの分解は、従来考えられている機構とは異なり、N末部のセリン残基の燐酸化には依存するが、N末部のユビキチン化部位には依存しないプロテアソーム蛋白分解機構によって起こると考えられた。現在我々は、ユビキチン化部位に依存しないプロテアソーム蛋白分解機構の解析を進めている。
