ダツクB型肝炎ウィルス(DHBV)の宿主レセプターgp180について以下の知見を得た。(1)DHBV初期感染におけるgp180の役割を理解するため、gp180上のDHBVpreS結合領域を決定した。ヒト・ダックgp180キメラを各種作製し、DHBVpreSとのbinding assayを行った。その結果、すでにニワトリ・ダックgp180キメラとのbinding assayから得られていたC末端側サブドメイン内の結合領域の他に、N末端側カルボキシペプチダーゼドメインに結合領域があることがわかった。(2)ダックgp180ドメインCのC末端側サブドメイン内のアミノ酸Tyr1250が、DHBVpreSとの結合に重要である。アルカリフォスファターゼ等の処理によってもgp180のpreSに対するアフィニティに変化はないことからこのアミノ酸残基への修飾は、preS結合に関与しないと結論した。(3)ダックgp180を強制発現させた293T細胞の粗抽出液を用い、多量のDHBVpreS存在下、FA-Ala-Lysを基質としてカルボキシペプチダーゼの活性を測定した結果、DHBVpreSの結合は、gp180の酵素活性に影響を与えないことがわかった。また、DHBV感染に関わる第二の宿主因子をダック肝臓cDNA発現ライブラリィより単離するための簡便で効果的な方法として、セレクティブマーカーとなるGFP DNAをウィルスゲノムに組み込んだDHBVウィルスベクターを構築した。
