Note (General): 金沢大学医薬保健研究域薬学系
吉田らの方法に準拠して、V1-ATPaseの回転の観察を行うために、遺伝子操作によりローターサブユニットである、ガンマsubunitにシステイン残基を導入したmutantV1を合計10種類作成した(ガンマサブユニット2,32,58,90,95,100,116,120,144,163のセリン、もしくはアラニンをシステインに置換)。蛍光化されたアクチン線維をローターサブユニットに結合させるためには、ガンマのシステイン残基をNEM-ビオチン化する必要がある。作成したmV1うち明確にビオチン化されるのは100番目のものだけであった。現在、このmutantV1を用いて回転の観察をおこなっている。いまのところ、明確な回転を観察できていない。この実験と平行して、ガンマsubnitの回転と共役したc-subunit oligomerの回転の観察も試みている。そのためにVoV1 operonのクローニングを行い、全塩基配列の決定と、構成subunitの決定に成功した(JBC274,inpress,on May)。現在、VoV1の発現系の構築を試みている。
