悪性神経膠腫の治療予後は極めて悪く、その病態の究明が必要である。我々は先行研究において悪性神経膠腫のエクソソーム内WT1が腫瘍の進展に関わっている可能性を示唆した。しかし、エクソソーム内WT1は既知のWT1と比較して分子量が高く、我々はSUMO化修飾によってWT1がエクソソームに移行するという仮説を立てた。結果、SUMO過剰発現神経膠腫株においてSUMO化WT1の発現上昇は確認できたが、SUMO化によるWT1のエクソソーム移行性の変化は見られなかった。SUMO化以外の修飾によるWT1のエクソソーム内移行の可能性を模索したが、エクソソーム内WT1の回収に難渋し、その究明には至らなかった。
The prognosis of malignant glioma is extremely poor, and its pathogenesis needs to be clarified. In our previous study, we suggested that WT1 in exosomes of malignant glioma may be involved in tumor progression. However, the molecular weight of WT1 in exosomes is higher than that of known WT1, and we hypothesized that WT1 is transferred to exosomes by SUMOylation.Although we observed increased expression of SUMOylated WT1 in SUMO-overexpressing glioma cell lines, we were unable to observe any change in translocation of WT1 by SUMOylation. We explore another modification which promotes WT1 translocation to exosomes, however it was difficult to collect large amount of WT1 in the exosomes and we failed to find this.
