【方法および効果】ヒト線維肉腫細胞のcell line(HT-1080)はheterogeneityを有している。Fidlerの方法に準じてin vivo selectionを行い10倍肺転移能の高いHT-1080のsubpopulationを得た。このsubpopulationは親株に比べてPAI-1の発現(ELISA)は3倍に増加していた。また限界希釈法を用いてHT-1080からPAI-1の発現が極めて少ないクローン(1-3C)と、それの多いクローン(26-6)を確立した。1-3CのPAI-1の発現量は親株HT-1080のわずか5%であり、肺転移形成能も低い。26-6のPAI-1の発現量は親株の約2倍で肺転移形成能は10倍以上であった。PAI-1の発現が少ない1-3CにPAI-1のcDNAを発現クローニングベクター(pcDNA1neo)に組み込み、リン酸カルシウム法、Lipofection法を用いてtransfectionを行った。neomycinによる選別を行いPAI-1がtransfectionされたcell line 1-3C(PAI)を得た。1-3C(PAI)は1-3Cの3倍以上のPAI-1の発現がみられた。また肺転移能に関しては、1-3Cはほとんど肺転移を形成しないにもかかわらず、1-3C(PAI)は高頻度に肺転移を形成した。【結論】PAI-1の発現が低いHT-1080のクローン(1-3C)にPAI-1のcDNAをtransfectionすることによりPAI-1の発現が増加し、さらに肺転移能の増加がみられた。PAI-1が肺転移を増加させる重要な因子の1つであることが直接的に証明された。
