1.細胞表層Fas抗原は細胞へapoptosisのシグナルを伝達する膜蛋白分子であるが,悪性グリオーマにおいてもその発現が報告されている。悪性グリオーマにおけるモノクローナル抗体である抗Fas抗体を用いたapoptosis誘導療法の可能性につき検討した。2.(1)C6ラットグリオーマ培養細胞の免疫組織学的検討においては,細胞膜にFas抗原の発現を認めた。(2)C6ラットグリオーマ培養細胞に抗Fas抗体(cloneCH11)100ng/mlを作用させたところ,72時間で75-95%の細胞がapoptosisをきたした。電顕とin situ nick endlabelling法での詳細な検討でも典型的なapoptosis像が観察された。(3)ラットC6脳内移植モデルを作成し,抗Fas抗体(cloneCH11)1000ng/mlを定位的に局所投与した。腫瘍内に細胞死を認めた。(4)ラット脳内(Wister Rat,6週齢)に抗Fas抗体(cloneCH11)を1000ng/ml注入し,3日目および7日目の組織を検討したが,ラットは全例生存し,特に神経学的異常はきたさず,組織学的にも大きな変化は認められなかった。(5)抗Fas抗体,抗Bcl-2抗体を使用し,ヒト剖検脳,ラット正常脳に対して免疫組織学的検討を行った。Fas抗原はラット腫瘍浸潤部のneuronやヒト照射脳のneuronの細胞膜に発現し,これらの細胞質にはBcl-2蛋白の発現も見られた。3.抗Fas抗体はC6グリオーマ細胞にapoptosisを誘導する。抗Fas抗体の脳内局所投与においてはneuronは細胞死を免れ,致死的合併症は認められなかった。bcl-2のapoptosis抑制効果の関与が考えられた。悪性グリオーマにおけるapoptosis誘導療法の可能性が広がった。
