EGFやPDGFなど増殖因子の受容体はリガンドの結合にともないエンドサイトーシスされリソソームで分解されるが(脱感作)、このとき受容体の細胞質領域がユビキチン化を受けることから、ユビキチンがエンドサイトーシスおよびリソソームへの輸送に関与する可能性がある。我々は野生型のユビキチンおよび、ジーロイシン・シグナル部分をアミノ酸置換した変異ユビキチンとインバリアント鎖とのキメラ蛋白をHeLa細胞に発現させ、そのエンドサイトーシスおよびリソソームへの輸送を検討した。その結果、変異ユビキチン・キメラでは野生型ユビキチン・キメラに比較してエンドサイトーシスは有意に遅延した。またエンドサイトーシスの後、野生型ユビキチン・キメラは比較的早期にリソソームに蓄積するのに対し、変異ユビキチン・キメラは1時間が経過した後もリソソームへは達しなかった。したがって、ユビキチン内のジーロイシン・シグナル配列は、ユビキチン化された膜蛋白質のエンドサイトーシスおよびリソソームへの輸送に関与すると考えられる。一方、温度感受性ユビキチン化変異CHO細胞へのヒトEGF受容体遺伝子導入に関しては、種々の変異を導入したEGF受容体cDNA発現ベクターを構築し、現在その安定発現株を作成中である。
