- 2009-2011年度科学研究費助成事業(科学研究費補助金(基盤研究(C)))研究成果報告書 課題番号:21580250 研究代表者:小松正治 (鹿児島大学水産学部准教授)マイクロシスチン-LR(MCLR)が示す細胞毒性と細胞増殖活性のバランス制御にp53が重要な機能を果たしていることを明らかにした。また、柑橘類のフラボノイドの一種ナリンジンが、OATP1B1/1B3を介したMCLRの細胞内取り込みを阻害することにより、細胞毒性の発現を抑制することを明らかにした。また、アメリカオオアカイカ由来のセラミドアミノエチルホスホン酸および海綿から単離したステリフェリンAにMCLRの細胞毒性を減弱する効果があることを突き止めた。This study demonstrates the importance of p53 in the regulation of cell fate after exposure to microcystin-LR. Our results suggest that, under conditions of p53 inactivation (including p53 mutation), chronic exposure to low doses of microcystin-LR may lead to cell proliferation through activation of Akt signaling. In addition, several compounds including naringin from grapefruit, stelliferin A from sponge, and ceramide aminoethylphosphonate from Hunboldt squid possessed the potent attenuation activity to the cytotoxicity of microcystin-LR. Results of this study may contribute to the development of chemoprevention and chemotherapeutic approaches to microcystin-LR poisoning.
