Note (General): 【背景】がんに対する治療法には, 一般的に外科手術, 化学療法, 放射線療法がある. その中でも化学療法は, 現在までにがんに対して優れた治療効果をもたらしてきたが, 抵抗性を示す場合も報告されている. また, 高濃度の抗がん剤は, 様々な副作用を引き起こすため, 現状よりも低濃度での使用が望まれている. そのため, 高濃度の抗がん剤と同様の効果を示すには, 他の補助療法が必要とされている. 一方, 腫瘍細胞で選択的に増殖する腫瘍溶解ウイルス(Oncolytic virus)によるがん治療は, 高い治療効果と選択性が期待される.口腔病理病態学教室では, アデノウイルスの複製に最も重要な E1A 遺伝子にAU-rich element（ARE）を組み込んだ腫瘍溶解アデノウイルス(Ad-ARET)を作成した. 本研究では, がん細胞に対する Ad-ARET とシスプラチンとの併用療法の効果を検討した.【材料と方法】ヒト腫瘍細胞 HeLa, SAS, A549, もしくはシスプラチン耐性を持つ SAS 細胞を用いて, Ad-ARET 単独, シスプラチン単独もしくは両者併用で細胞を処理した. これらの細胞の生細胞活性を MTS アッセイで検討することにより細胞死を検討し, また, ウイルスの増殖も解析した. さらに, シスプラチン処理により, ARE 結合タンパク HuR の局在を検討することにより, ARE-mRNA の安定化およびウイルスの増殖を解析した. 【結果】いずれの細胞でも, Ad-ARET あるいはシスプラチン単独処理よりも, 併用処理の方が, がん細胞死活性が高かった. また, シスプラチン存在下で, HuRの細胞質局在が認められ, ARE-mRNA の安定化により Ad-ARET の生産効率が上がることが解明された.【結語】Ad-ARET はシスプラチンと併用することにより, より高い腫瘍細胞溶解効果を持つことが示された. これは, シスプラチン処理によって, HuR タンパクが細胞質に核外輸送され, ARE-mRNA が安定化されることにより, Ad-ARET がより増加したことによると考えられる. 以上より, Ad-ARET とシスプラチンとの併用療法は有効な治療法になりうると思われる.
Oncolytic adenovirus is an attractive tool for cancer therapy. We developed a new oncolytic adenovirus Ad-ARET including AU-rich element (ARE) in the 3’-utranslated region (3’-UTR) of E1A gene. ARE is the RNA element which enhances the rapid decay of mRNAs including those of the genes required for cell growth and proliferation. In the vast majority of cancer cells, the amount of cytoplasmic HuR, which binds to ARE and protects ARE-mRNA from rapid degradation, up-regulated and ARE-mRNA is stabilized. And the stabilization is co-related with the malignancy of the cancer cells.Cisplatin has a significant anti-tumor activity against a wide variety of tumors. Since cisplatin may injure normal cells, ineffective drug treatment to cancer tissue causes the side effect. The combination of oncolytic virus and cisplatin can allow the use of reduced dosage of each reagent, and reduce side effect. In this study, I examined the combination effect of Ad-ARET with cisplatin using several kinds of cancer cells. In the case of combined use of Ad-ARET with cisplatin, the cell survival rate of cancer cells were lower compared with those of single treatments. The combined effect was also evident in cisplatin-resistant cells. I found that cisplatin treatment induced HuR export and resulting in up-regulation of Ad-ARET replication through E1A mRNA stabilization. These results indicate that Ad-ARET is available for the combination cancer therapy with cisplatin.
