Note (General): ヒト脱落乳歯歯髄幹細胞(SHED)は, 乳歯歯髄組織に存在している間葉系幹細胞であり, SHED が放出する分泌因子には, 組織修復能と免疫調整能があることが報告されている. 本研究は, 骨粗鬆症に対するSHED の有効性について検討を行った.交換期のため脱落した乳歯からSHED を分離し, 継代培養を行った後に, 培養上清を回収し実験に用いた. 11 週齢C3H/HeJ 雌マウスに対し卵巣摘出(OVX)を施行, OVX 直後から週2回, 無血清培地(SF-MEM 群)あるいはSHED 培養上清(SHED-CM 群)を腹腔内に投与した.投与開始4 週後に, 大腿骨の状態をμCT で観察した. さらに, 腹腔内液におけるマクロファージ分画と, 末梢血中のIL-17 およびIFN-γ 産生細胞数についFACS 解析を行った.大腿骨における骨量は, SHED-CM 群ではSF-MEM 群と比較し有意に高い値を示した. IL-17 およびIFN-γ 産生細胞数は, SHED-CM 群では有意に減少していた. また腹腔内液中のマクロファージは, SHED-CM 群ではM2 マクロファージの有意な増加を認めた. さらに, 中和抗体を用いてM2 マクロファージの発現を抑制したところ, SHED-CM の投与にも関わらず,IL-17 およびIFN-γ 産生細胞数と骨量は, SF-MEM 群と有意差は確認されなかった.OVX による骨粗鬆症において, SHED-CM は, 骨吸収抑制効果を有することが明らかになった. このSHED-CM による骨吸収抑制効果は, M2 マクロファージとIL-17 およびIFN-γ産生細胞の活性が関わっていることが示唆された.
