Note (General): 【目的】Mineral trioxide aggregate（MTA）にリン酸化プルランを添加した試作根管充填用シーラーは、高アルカリ性を維持しCa2+の溶出に優れ，高い封鎖性を有することが報告されている．本研究の目的は，試作シーラーを用いて根管充填した後の，根尖孔へのセメント質様硬組織の形成と根尖性歯周炎の治癒に及ぼす効果を組織学的に評価することである．【材料と方法】まず試作シーラー（TS），AH プラス（AH），MTA フィラペックス（MF），およびNEX MTA セメント（NM）からのCa2+溶出量を12 週間測定した．次にビーグル犬のP1～P4 を抜髄し， TS，AH，MF とガッタパーチャポイントを用いて根管充填を行い2，6 週後に根尖孔のセメント質様硬組織による封鎖率を組織学的に計測した．さらに，ビーグル犬のP1～P4 を抜髄，根管内にプラーク懸濁液を封入して根尖性歯周炎を誘発した．4 週後， TS，AH，MF とガッタパーチャポイントを用いて根管充填し、2，6 週後に根尖部骨欠損面積を組織学的に計測するとともに，根尖孔のセメント質様硬組織による封鎖状態を評価した．【結果と考察】TS のCa2+溶出は12 週まで持続し、溶出量はNM，TS，MF の順で多く，TS はMF の1.25 倍，NM の0.83 倍であったが、AH からは検出されなかった．また，抜髄後に根管充填を行った歯根では、2，6 週後とも硬組織による根尖孔封鎖率はAH，MF に比べてTS が有意に高い値であった．一方、根尖性歯周炎誘発して根管充填を行った歯根では、6 週後、TS の骨欠損面積はAH， MF より有意に小さく、硬組織による根尖孔封鎖率はTS が他の2 群に比較して有意に高い値を示した．これらの結果から，TS は歯内療法の成功率を向上させると思われた．【結論】リン酸化プルラン含有MTA シーラーは，AH プラスとMTA フィラペックスに比較して硬組織による根尖孔封鎖性に優れ、根尖性歯周炎の治癒促進効果を有することが示された．
