• Note (General): これまでの研究で、硫酸化糖鎖の抗ウイルス性はHIVやインフルエンザウイルスの表皮タンパク質と硫酸化糖鎖との静電的相互作用、すなわち、硫酸基に由来する(-)電荷とウイルス表皮タンパク質中の(+)電荷との静電的相互作用によると考えられている。本論文の第1章は序論、第2章では抗ウイルス性バイオ材料の合成を目的にβ-シクロデキストリン（β-CD）の6位にClick反応を利用してオリゴ糖鎖を結合させた新しい硫酸化β-CD糖鎖誘導体を合成し抗HIV性について検討した。第3章では硫酸化アルキルオリゴ糖鎖の高い抗HIV性を解明する目的でリボソームとの相互作用をSPRを用いて検討した。第4章はまとめである。 第2章で、β-CDの6位水酸基をヨウ素化しNaN3を用いた求核置換反応によりアジド基を導入し、続いてグルコースの1位をプロパギル化しアジド化β-CDとのClick反応を行った。脱アセチル化、硫酸化してMTT法により抗HIV性を測定した。マルトースからも同様に合成した。FT-IR、高分解能NMRなどを用いて構造解析を行った。1H NMR測定から4.5ppmの6位水酸基プロトン（6-DH2OH）シグナルが完全に消失し、またFT-IR分析で新たに2108cm-1にアジド基に由来するシグナルが現れたことにより7コの水酸基はすべてアジド基に置換されたことが分かったClick反応も効率的に進行しβ-CDの6位にトリアゾール環を介してグルコースまたはマルトースが導入された。抗HIV性は硫酸化β-CDではほとんど示さなかった（EC50>200μg/mL）が、グルコース分枝では27.9μg/mL、マルトース分枝では1.3μg/mL、50％細胞障害濃度（細胞毒性）はCC5>200μg/mL（デキストラン硫酸ではEC50=0.3μg/mL、CC50>1000μg/mL）となり、分枝糖鎖が大きくなると高い抗HIV性を示した。抗ウイルス性は分子量に依存するが、β-CDにグルコースなどを分枝させるとクラスター効果により高い抗HIV性を示したと考えた。 第3章で、続いて硫酸化アルキルオリゴ糖鎖の高い抗HIV性を解明するために硫酸化アルキルマルト7糖を合成しリポソームとの相互作用を測定した。その結果、リポソームの脂質二重膜にアルキル鎖が相互作用することを見出した。この結果は長鎖アルキル鎖を導入した硫酸化糖鎖とリポソームとの相互作用と同じ結果であったことから、硫酸化アルキルオリゴ糖鎖の長鎖アルキル鎖がHIVの脂質二重膜と相互作用して抗HIV性は発言するものと考えた。; We have synthesized sulfated polysaccharides with potent ant-HIV activity by the two methods as following. The first one is ring-opening polymerization of protected anhydro sugar monomers and deprotection followed by sulfation, and the second one by sulfation of naturally occurring polysaccharides. It is for investigation of biological activity of poly and oligosaccharides to make a strategic elucidation both synthesis and natural products. On the other hand, it is interested in the functionality of oligosaccharides. Sulfated oligosaccharides have low cytotoxicity, but, low anti-HIV activity. Therefore, lysine dendrimers connected with sulfated oligosaccharides were found to give high anti-HIV activity by making use of cluster effect of oligosaccharides. It was reported previously that sulfated oligosaccharides with high anti-HIV activity was synthesized by introduction of long-chain alkyl group at the reducing terminal. In this thesis, I easily synthesize oligosaccharide derivatives by using click reaction as follows, and anti-HIV activity and mechanism were investigated. Click reaction with 1-O-propagyl maltose and 6-azido-6-deoxy β-CD was carried out to give oligosaccharide-branched β-CDs at C6 position through triazol ring and then sulfation. I found that the sulfated oligosaccharide-branched β-CDs were found to have potent anti-HIV activity even though sulfated β-CD had low anti-HIV activity. The exact results and proposed anti-HIV mechanism are described in Chapter 2. In addition, previous reports indicated that the introduction of a long-chain alkyl group at the reducing end of oligosaccharides and subsequent sulfation was enhanced anti-HIV activity. However, the role of the long-chain alkyl group is still unclear. In Chapter 3, I focused on the role of the long-chain alkyl group by investigating the interaction with liposome as a model of HIV. Sulfated alkyl oligosaccharide which was prepared by click reaction between 1-azide-1-O-deoxy-acetylated maltoheptaoside and 1-decyne and then sulfation. The interaction with liposome is elucidated by using a surface plasmon resonance (SPR) and dynamic light scattering (DLS) measurements. In this thesis, a new utilization of oligosaccharides is exhibited by cluster effect of oligosaccharides with β-CD core and the role of long-chain alkyl group in sulfated alkyl oligosaccharides to enhance anti-HIV activity is described. I showed a new direction for using oligosaccharides on antiviral activities.
