Note (General): ヒト脂肪由来幹細胞をストロンチウム(Sr)含有軟骨形成培地で培養すると、軟骨分化が促進されるか調べた。健康被験者の頬部脂肪体から脂肪組織を採取し、初代培養を行った。フローサイトメトリーにより脂肪由来幹細胞(ADSC)を単離し、基礎培地のみ、軟骨分化培地(CM)、Sr添加CM培地のそれぞれで培養した。軟骨形成はアルシアンブルー染色により調べた。培養細胞からtotal RNAを抽出し、逆転写反応によりcDNAを合成し、PCR法を用いてmRNAの発現を調べた。1.50mM Sr含有CMでは基礎培地のみとCMのみの場合と比べて、グリコサミノグリカン分泌と、SRY(sex determining region Y)-box 9およびcollagen type 2 alpha 1のmRNA発現が有意に高かった。脂質、骨芽細胞、肥大性軟骨への分化マーカーの発現増加は認めなかった。SrはADSCの軟骨分化を誘導する可能性が示唆された。
