Note (General)Orthodontic tooth movement is based on the response of biological tissue to mechanical loading. On both the compressed side and tensile side, stress is loaded onto cells by means of physical stimulation that causes cells to produce various cytokines that are involved in the promotion or suppression of osteoclast formation. During orthodontic tooth movement, alveolar bone resorption at the area of compression occurs through osteoclastic activity. In this study, we confirmed the coordinated effect of IL-17A and IL-27 on receptor activator of nuclear factor kB ligand (RANKL)-induced osteoclastogenesis, using RAW264.7 cells as osteoclast precursors. High expression of IL-17RC and IL27-Rα was detected on RAW264.7 cells, and this expression was not altered by the stimulation of RANKL for 24 h. A coordinated effect of IL-17A and IL-27 suppressed osteoclast differentiation from RAW264.7 cells treated with RANKL without reduction of cell proliferation. Furthermore, the phosphorylation of c-Jun-N-terminal kinase (JNK) stimulated with RANKL was inhibited by the coordinated effect of IL-17A and IL-27. These data suggest the possibility that the coordinated effect of IL-17A and IL-27 inhibited the phosphorylation of JNK which is involved in the suppression of RANKL-induced osteoclast differentiation in RAW264.7 cells. Our findings provide new insights into the mechanisms of osteoclastogenesis.
