Note (General): Cancer stem cells (CSC), which play important roles in tumor initiation and progression, are resistant to many therapies. However, the regulatory mechanisms underlying CSC-specific properties, including self-renewal, are poorly understood. We evaluated the immunohisto-chemical expression of NF-κB and CD44, which is also considered an indicator of CSC, in oral squamous cell carcinoma (OSCC) clinical samples. Moreover, we analyzed the effects of NF-κB inhibition on epithelial-mesenchymal transition (EMT) and migration using western blot analysis and knockdown by NF-κB inhibitor BMS-345541 in OSCC. Immunohistochemical staining of NF-κB and CD44 was strongly expressed in the invasion front of the tumor cells.　In OSCC cell lines, expression of NF-κB was observed in HSC3, HSC4 and KB, and in particu-lar, phosphorylated NF-κB was clearly expressed in KB, showing stem cell-like properties. Inhibition of NF-κB by BMS-345541 effectively inhibited the expression of protein involved in the EMT, and the migration of OSCCs. These results suggest an important role of NF-κB in oral cancer EMT and metastasis-related migration. NF-κB-targeted treatment may be an ef-fective therapy for metastasis of oral cancer.
