Note (General): [要旨]Wntは分泌性の糖タンパク質であり，近年Wntシグナルが骨形成に重要であることがわかってきた．歯周組織の維持や改造にもWntシグナルが関与することが報告されている．しかし，生理的状態において，Wnt/β-cateninシグナル陽性細胞が歯根膜を含む歯周組織のどこに分布するか，また，その分布が加齢により変化するかどうかは未解明な部分が多い．本研究では，Axin2CreERT2-tdTomatoマウス(4週齢，6週齢，30週齢，48週齢)を用いて，歯周組織におけるWnt/β-cateninシグナル活性化細胞を観察した．Wnt/β-cateninシグナルの活性化は，その標的遺伝子であるAxin2が発現すると赤色蛍光タンパク質であるtdTomatoが発現することを指標にした．タモキシフェン(TAM)投与後3日において，Axin2の発現は，4週齢および6週齢では根尖部歯根膜，根管歯髄，根間中隔部歯槽骨および歯肉上皮部に多く認められた．しかし，30週齢マウスでは，Axin2発現細胞は減少しており，48週齢マウスでは，その発現はわずかであった．TAM投与後30日では，Axin2の発現細胞がすべての週齢において増加したものの，48週齢マウスではAxin2発現細胞の増加をわずかに認めるのみであった．Axin2発現細胞は，細胞増殖マーカーであるKi67を発現した．つまり，Axin2発現細胞は細胞増殖活性が高いことを示唆する．加齢に伴いAxin2陽性かつKi67陽性細胞は減少した．一方，加齢に伴い，歯槽骨2におけるSclerostin陽性骨細胞が増加した．以上よりWnt/β-cateninシグナル陽性細胞は，歯根膜，歯肉上皮，歯槽骨といった歯周組織や歯髄に分布し，細胞増殖が旺盛であることが示唆された．また，加齢に伴い，歯骨でのSclerostin発現が増加するため，Wnt/β-cateninシグナル陽性細胞が減少する可能性が示唆された．若齢マウスでは，根尖部歯根膜と根尖部歯髄象牙芽細胞でWnt/β-cateninシグナルが強く活性化されていた．加齢に伴い，Wnt/β-cateninシグナル陽性細胞とその細胞増殖は顕著に減少した
