Note (General): 活性化転写因子1（ATF1）は、CREB/ATFファミリーの転写因子に属し、精巣で高発現している。しかし、精子形成におけるATF1の役割は未だ確立されていない。本研究では、精子形成におけるATF1の影響を解明することを目的として、マウスにおけるATF1の発現パターンとマウス精巣におけるATF1ノックダウンの影響を調べた。その結果、ATF1はさまざまな臓器で発現しており、精巣では非常に高いレベルで発現していることがわかった。免疫染色により、ATF1はspermatogoniaの核に局在し、増殖細胞核抗原(PCNA)と共局在することがわかった。ATF1欠損マウスでは、精巣の精細管にはすべての発生段階の細胞が存在していたが、spermatocyte以降の分化段階の細胞数は減少していた。同様にPCNAの発現が低下していた。一方で、精細管におけるアポトーシス細胞はほとんど見られなかった。これらの結果は、ATF1が男性生殖細胞の増殖と精子の生成に関与していることを示している。また、男性不妊症における乏精子症、無精子症の発症機序解明の可能性を示唆した。
