Note (General): Retinal vasculopathy with cerebral leukoencephalopathy and systemic manifestations (RVCL-S)はエキソヌクレアーゼ活性を持つthree-prime repair exonuclease 1(TREX1)の遺伝子変異による常染色体優性遺伝疾患である．成人期に脳小血管病変を来し，10年ほどで致命的な転機をたどる．RVCL-Sを引き起こす変異ではエキソヌクレアーゼ活性を維持しているが，野生型で細胞質中の小胞体膜に局在しているTREX1が核の内部にも存在することが知られている．今までにRVCL-Sの動物モデルとして複数の試みがされているが，TREX1の変異がRVCL-Sの分子病態にどのように関与しているのかは不明である．そこで，RVCL-Sの病態機序解明のためには，より扱いやすい動物モデルの構築が望ましい．今回，RVCL-Sで頻度の多いp.Val235GlyfsTer6(hTREX1 V235fs)変異TREX1を発現させるためのショウジョウバエモデルを作製した．hTREX1 V235fsを発現したハエでは，TREX1タンパク質は核を含む細胞全体でびまん性の局在を示した．また，野生型と比較して複眼の形態異常が有意に生じた．さらに，ハエ全神経細胞やハエグリア細胞にhTREX1 V235fsを発現した場合と比べてハエ気管に発現した際に毒性が高く、変異型TREX1の細胞毒性は組織特異性を有する可能性が示唆された．以上の実験によって，RVCL-S変異ヒトTREX1発現ショウジョウバエモデルを樹立した．本モデルはRVCL-S病態解析のための遺伝子スクリーニングや薬剤スクリーニングに利用することが期待できる．
