Note (General): Pentraxinは免疫反応に重要な役割を果たす5量体の蛋白で、pentraxin 3 (PTX3)やC-reactive protein (CRP)はpentraxin superfamily に属する。pentraxin 3 (PTX3) は炎症性刺激に曝されるとマクロファージ、好中球、血管内皮細胞など様々な細胞から産生され、好中球ではPTX3は細胞質内の顆粒に局在する。好中球の顆粒は1次顆粒、2次顆粒、3次顆粒の3種に分けられ、 PTX3 は2次顆粒（特殊顆粒）に存在すると考えられてきた。しかし、他の顆粒における局在については不明であった。本研究では成熟好中球と好中球へ分化誘導した前骨髄球性細胞株HL-60細胞を検討し、すべての顆粒にPTX3が存在することを確認した。IL-8の刺激によって好中球はneutrophil extracellular traps (NETs) と呼ばれる不規則な網状形態を示したが、3種の顆粒はこのNETsの表面に接着し、PTX3はこれら顆粒と共局在した。以上の成績からPTX3は好中球の細胞質内および細胞外の顆粒構成抗菌性蛋白と共存し、生体防御上、重要な役割を果たしていることが推測された。
