Note (General): コンブの一種Saccharina japonicaは、北海道を中心とする北太平洋沿岸西部の寒海域に広く分布し、各地で大規模な藻場を形成する生態的な重要種である。本種は形態変異に富み、その生育環境もさまざまであるが、近年の地球規模の環境変化によって北日本沿岸で広く資源量は減少している。今後、本種在来個体群の遺伝的固有性を考慮した藻場造成を行うために、集団構造を詳細に理解し、適切にその多様性の保全に努める必要がある。本研究では、北海道沿岸とその周辺域のS.japonica について、個体群間の遺伝的多様性をDNA多型解析によって明らかにするため、先ずは、粘質多糖類を多量に含む葉体から解析に有用な高純度DNAの抽出方法の確立を試みた。そのなかで今回、組織の洗浄、SDS処理、CTAB処理を作業の柱とする方法が見出され、得られたDNAを用いたAFLP分析では高い再現性のある結果が得られた。また、抽出DNAを鋳型としたDual-Suppression PCR法によりマイクロサテライトマーカー10種類も開発することができた。AFLP分析とマイクロサテライトマーカー分析の結果、本種内の既存変種の枠組みを超えた地域個体群の存在が認められたほか、周辺の地域とは遺伝的特性の異なる孤立した遺伝子集団の存在も明らかになった。さらに、単一産地内の群落についても物理的環境要因によって遺伝的な分化が生じることが示唆され、このような小規模群落を維持ことも本種の遺伝的多様性を保全するうえで重要であることが示された。また、本研究では S.japonica 藻場の衰退に深く関わる環境因子である水温と栄養塩濃度の変化に依存して発現量が増減するタンパク質を探索し、vBPO などの環境ストレス応答タンパク質をマーカーとした藻場の健全性を計る技術を開発した。さらに、本種の小規模群落や、胞子の供給源となる核藻場の保全に向けて、コンブ葉体由来の揮発性化合物がコンブ摂餌動物（キタムラサキウニ）誘引に働くことを確かめ、化合物添加の高分子ゲルを利用した新たなウニの駆集方法を提案した。
