• Note (General): 各臓器由来のアルカリ性ホスファターゼ（ALP）の生体における真の基質と機能に関して未だに不明な点が多い．そこで，ヒトの骨型，小腸型，胎盤型及び肝臓由来のALPを購入して使用し，それらの基質選択性及び阻害剤に対する感受性を検索した．基質として，生体内には存在しないがALP活性の測定に汎用されるパラニトロフェニルリン酸（p-NPP）と，生体内に存在するナトリウムピロリン酸（Na-PPi），アデノシン三リン酸（ATP）及びピリドキサルリン酸（PLP）を使用して，各ALP活性のpH依存性と基質濃度依存性、及び阻害剤であるlevamisole及びvanadateによる阻害を検討した．活性は各基質が加水分解されて生ずる無機リンをChifflet法によって定量して測定した．骨型ALPの生体内基質とされているピロリン酸分解の至適pHはどのALPも8.8から9.1であり，また50%活性化濃度（Ko.5）もALPの種類によらず3から5mMであった．またピロリン酸を基質とした際の阻害剤の50%活性阻害濃度（Kio.5）も類似しており，ALPの種類による違いは見いだせなかった．小腸型ALPの基質とされるPLP分解の至適pHはどのALPも9.9から10.4であり，またPLP分解のKo.5もALPの種類によらず1.1から1.6mMであった．PLPを基質とした際の阻害剤のKio.5は，vanadateではALPの種類によらず類似していたが，levamisoleでは異なっていた．ATPあるいはp-NPPを基質として同様の実験を行った結果，基質に対する至適pHと基質に対するKo.5はALPの由来には関係なく基質によって類似していた．また，levamisoleなどの阻害剤に対するKio.5は基質の種類には関係なく，ALPの種類によってほぼ一定であった．ヒトに存在する各臓器由来のALPはその機能と生体内基質に対応して変化してきた可能性があると考えて本研究を行ったが，少なくとも今回検討した基質に関してはALPの種類による相違は見いだせず，むしろ阻害剤に対する感受性にALPの種類による相違が見られた.; The human alkaline phosphatase(ALP) is homologous to not only other mammal ALP but also bacterial ALP. However, the substrate and function of ALP in vivo have not been cleared. Recently, we obtained human ALP as a chemical reagent. In this study, we examined the difference in substrate selectivity and inhibitor sensitivity among several types of human ALP. We used 4 types of human ALP from bone, intestine, placenta or liver. p-nitrophenyl phosphate(p-NPP), sodium phyrophosphate(Na-PPi), adenosine triphosphate(ATP) or pyridoxal phosphate(PLP) were used for substrate. The amount of inorganic phosphate as a result of hydrolysis by ALP was measured by Chifflet methods under different pH or substrate density. To examine the inhibitory effect on activity, levamisole or vanadate were used. The substrate density exhibiting 50% activity and the density exhibiting 50% inhibition were proposed as Ko.5 and Kio.5 respectively. When the Na-PPi, the substrate of bone ALP, was used, the optical pH and Ko.5 did not show differences among each ALP types (optical pH : 8.8 ~ 9.1, Ko.5 : 3 ~ 5 mM). In case of PLP, the substrate of intestine ALP, the optical pH and Ko.5 did not show differences among each ALP types(optical pH : 9.9 ~ 10.4, Ko.5 : 1.1 ~ 1.6 mM), although the optical pH sifted to alkaline side. Additionally, the optical pH and Ko.5 on ATP or p-NPP, it is not in human body but many researchers used, did not show differences among each ALP types. However, in case of PLP, Kio.5 of levamisole was different among each ALP types, although Kio.5 of levamisole or vanadate did not show any differences among each ALP types.
