• Note (General): 水稲栽培において浸種は， 発芽には至らない15℃以下の水温で発芽開始前の吸水条件を揃え，また発芽抑制物質を取り除くことにより，斉一かつ良好な催芽種子を得ることを目的とした栽培体系の第一段階に行われる作業であり，重要な栽培管理として古くから一般的に行われている．しかし，日本の水稲生産において最も水稲栽培の作付面積の多い地域である新潟県では，初春に開始する種子予措は育苗施設の状況により屋外での浸種も行われているが，浸種を開始する3月末から4月前半は上水道の水温は低く，浸種期間の外気温も5℃やそれ以下の低温になる場合があり，その場合しばしば発芽不良の問題が生じている．新潟県の種子生産に用いられる原種は，各種子場に配付する前に原則として形質異常，混種が認められないことを実際に確認栽培した後に配付されるため， 種子場では長期貯蔵種子（1年以上貯蔵した種子） を用いた育苗が主に行われている． 新潟県で作付けされる主要品種のうち， その約70%の作付面積を占める主食用品種のコシヒカリは，長期貯蔵種子においても低温浸種の発芽率への影響は比較的小さいが，新潟県の主要品種の一つである糯品種のこがねもちや酒米用品種である五百万石は，発芽能力の低下が顕著である．新潟県内で報告された過去の発芽不良の原因は，催芽時間の不足といった人為的ミスの他，主にこれらの品種で浸種温度が低い場合に発芽不良が生じているケースが多い．そのため，こがねもちや五百万石等の長期貯蔵種子では浸種水温を10℃以上に保つ必要があるという知見に基づき（佐藤ら， 2003），各指導機関から情報提供が図られているが，未だ発芽不良が生じる場合がある．長期貯蔵種子の発芽不良の軽減には，浸種初期水温の確保が重要であり，三重県での事例とその再現試験では，浸種開始24 時間の水温を17.5℃ で保つことにより発芽不良が改善され，10℃以上の浸種処理と同様の発芽率が確保できることが報告されている（北野ら， 2010）． そこで，本学位論文では，新潟県で発芽不良問題を生じやすい水稲主要品種について種子の貯蔵期間， 浸種条件が発芽率に及ぼす影響をシャーレ試験により調査し，それに対応した種子浸種条件を検討した．その結果，長期貯蔵種子は，10℃以下で浸種すると無浸種のものよりも発芽率が低下し，この低水温浸種期間が長いほどさらに発芽率の低下を招くが，浸種初期1日目の水温を20℃に保ち，その後4日間を5℃で処理した場合，10℃以上で5日間の浸種処理と同様の発芽率が確保できることがわかった．さらに，種子生産現場において加温装置のない水槽でも導入可能な，浸種開始時の水温を高めることによる発芽障害の回避及び良好な出芽を得られる方法について，実際の浸種・育苗条件に近い屋外での再現実証試験を行い，浸種開始時における最適な水温条件とその効果を検討した．その結果，こがねもち及び五百万石の長期貯蔵種子は，浸種開始水温を30～50℃とすることで低温による発芽阻害を回避できることがわかった．; 現場の多様な状況に応じた，発芽不良を最小限に抑える浸種対策をより適確に対応させるためには，どのポイントが発芽不良の原因となるのか，そのメカニズムを把握することが必要である．そこで，浸種初期の水温の違いがなぜその後の発芽に影響を及ぼすのかについて詳細な検討を行うため，浸種初期における①低水温が発芽率に影響を及ぼす時間範囲，②水温を高くすることが発芽率に影響を及ぼす時間範囲，についてそれぞれ調査を実施した． その結果， 貯蔵1.5～ 2.5年の五百万石， こがねもち及びこしいぶきでは， 浸種開始24時間以内の水温が浸種後の発芽に最も重要な影響を及ぼすことがわかった．この間の浸種水温が5℃ではその後の水温が12℃ でも発芽率が低下する一方で， 浸種初期24時間の水温が12℃もしくは80分間が30℃の場合には，その後の水温が5℃でも，水温12℃で5日間処理した場合と同等以上の発芽率が得られた．さらに，低水温浸種による発芽率低下の内因的な発生要因を特定するため，低温浸種後の催芽時の種子におけるタンパク質発現を，イネの発芽において重要なデンプン加水分解酵素であるα-アミラーゼのアイソフォーム数種について，ウェスタンブロッティングによる発現解析で検証を行った． α-アミラーゼは各種アイソフォーム毎に異なる役割を持つことが知られているが（Mitsui et al., 1996），ここでの低水温浸種による影響もアイソフォーム毎に異なる発現傾向が確認された． α-アミラーゼI-1及びII-3では不発芽種子における発現低下が確認された一方で， α-アミラーゼII-4では不発芽種子で比較的強い発現が確認され， α-アミラーゼII-6では品種毎に異なる発現を示す等，アイソフォームによる発現の差異があることがわかった．以上，本学位論文では長期貯蔵種子の低水温浸種による発芽阻害要因について，特に浸種開始時における水温の差異がその後催芽時の発芽に重要な影響を及ぼし， また内因的にもα-アミラーゼの発現変動を引き起こしているという結果を得た．これらの結果は，水稲生産現場において低温に遭遇した場合等の環境下で，浸種処理で重視すべき点を把握する上で重要であり，その後の育苗に及ぼす影響を考慮して種子予措を行うことで，良質苗の安定確保を図ることができるものと考えられる．; Rice (Oryza sativa L.) seeds are soaked before germination to ensure uniformity of seedling growth by removing germination inhibitors (Mikkelsen and Sinah, 1961; Takahashi, 1967). In Japan, breeder seeds are stored for more than a year before planting to allow characteristics to be confirmed and to verify freedom from contamination. Seeds stored long term frequently produce stunted and uneven seedlings under some soaking conditions. The ideal temperature and time of soaking to ensure good germination and seedling emergence of long-term-stored seeds are not understood yet. Seeds are often soaked outdoors. In Niigata Prefecture, the outside air temperature in late March to early April can fall below 5 °C, lowering water temperature with it. The three main cultivars grown in Niigata—Koshihikari (nonglutinous rice), Koganemochi (glutinous rice), and Gohyakumangoku (brewers’ rice)—exhibit different susceptibilities to low soaking temperature. Koganemochi and Gohyakumangoku seeds stored long term must be soaked at >10 °C for good germination (Sato et al., 2003). It is empirically known that 100 °C•day is necessary to remove the effects of dormancy (Hirai et al., 2008). In addition, the correct initial soaking temperature is necessary to improve the germination of long-term-stored seed: germination was improved by an initial temperature of 17.5 °C (Kitano et al., 2010). However, it is still unclear why the initial water temperature influences germination. Here, we investigated the germination abilities of long-term-stored seeds of rice soaked at different temperatures for different times. The germination efficiencies of seeds soaked at 5 °C for 5 day were markedly less than those of seeds soaked at 12 °C for 5 day. Poor germination caused by soaking at 5 °C for 24 hour was not rescued by additional soaking at 12 °C for 4 day. On the other hand, the germination efficiencies of seeds soaked either at 12 °C for 24 hour or at 30 °C for 80 minute were similar to those of seeds soaked at 12 °C for 5 day, even when further soaked at 5 °C. Thus, the soaking temperature within the first 24 hour was extremely important for the germination of long-term-stored rice seeds.; It is widely accepted that α-amylase plays a key role in degrading the starch reserves in germinating cereal seeds (Akazawa et al., 1988; Fincher, 1989; Jones and Jacobsen, 1991; Asatsuma et al., 2005). Germination and seedling growth were markedly delayed in α-amylase-suppressed rice seeds (Asatsuma et al., 2005). Therefore, we also examined the expression pattern of α-amylase isoforms in germinating and non-germinating seeds of cold-soaking-tolerant and susceptible rice cultivars. Seeds stored for 2.8 years were germinated, and α-amylases were analyzed by western blotting with specific antibodies (Mi tsui et al., 1996; Nanjo et al., 2004a). The expression of AmyI-1 was high in the germinating seeds of all three cultivars, but low in the non-germinating seeds. The pattern of AmyII-3 expression in these seeds was similar to that of AmyI-1 expression. It is noteworthy that AmyI-1 expression level in non-germinating Koshihikari seeds was higher than those in Gohyakumangoku and Koganemochi. AmyII-4 expression was lower in the germinating seeds than in the non-germinating seeds. AmyII-6 expression was higher in the germinating seeds of Koshihikari than in the non-germinating seeds, but lower in the germinating seeds of Gohyakumangoku andKoganemochi than in the non-germinating seeds. In summary, we investigated the physiological and biochemical influence of cold-water soaking on seed germination. The soaking temperature within the first 24 hour was extremely important for the germination of long-term-stored rice seeds. Notably, soaking at 30 ° improved germination in soil under conditions similar to field conditions. We also found differences in expression patterns of AmyI-1, AmyII-3 and AmyII-6 in imbibed seeds between cold-soaking-tolerant and -susceptible cultivars.
