Note (General): 近年，超高齢社会となり高齢者のQuality of life（QOL）を低下させ得る骨粗鬆症による骨折が多くなってきているが，骨粗鬆症に関係する破骨細胞の分化誘導系に対し，重大な副作用なく効果を示す新しい薬物が望まれている．本研究では，RAW264.7 破骨細胞前駆細胞を用いて既に急性期の脳梗塞に対して臨床応用されているエダラボンが，RANKL誘導性の破骨細胞分化誘導系に与える影響について検証した．RANKL添加日を１日目とした場合，エダラボンの添加を１日目と３日目の両日（A群），１日目のみ（B群），３日目のみ（C群）と場合分けした時の総破骨細胞数，核数別破骨細胞数の計測を行った．また，Real-time-PCR 法によりエダラボン添加後の3，6，12，24，48時間後の破骨細胞関連遺伝子のmRNA発現量を計測した．50 μM エダラボンの添加により，総破骨細胞数はA，B 群では対照群と比較し約45%減少し，C群では約15%減少した．核数別の破骨細胞数は少数核の破骨細胞でも巨大破骨細胞でも有意な減少を認めた．Real-time-PCR法による破骨細胞関連遺伝子のmRNA発現量は，RANK，NFATc1，TRAP，Cathepsin K，OC-STAMPにおいて一時的な減少を認めた．以上からエダラボンの添加により破骨細胞分化関連因子が抑制されることにより，破骨細胞の分化誘導が抑制される可能性が示唆された．
